There is a growing trend in Western countries to use herbal medicine as alternative medicine to treat a wide range of diseases, e.g. chronic liver diseases. We have identified a Chinese herbal medicine for the treatment of liver cirrhosis and hepatocellular carcinoma (HCC). To develop this herbal medicine as therapeutic agent and to address many hurdles to place traditional Chinese medicine into mainstream medical care, we aim to address three challenges in herbal medicine research, namely lack of evidence for systematic scientific evidence, inconsistency of herbal quality, and the unknown mechanism of action. Here, we provide a solution for these challenges by using Connectivity Map, a database developed to store expression profiles of disease, gene, and chemical expression profiles. We have developed a pattern-match platform by using gene expression signatures from TCM-treated cells as queries to make inference on the connection among the query, internal profiles stored in connectivity map and collected HCC patient gene signatures. Moreover, we hypothesized that if a drug signature could reverse, at least in part, the gene expression signature of HCC, this drug might have the potential to inhibit HCC pathways and thereby treat HCC. Many drugs (small molecule inhibitors) have been validated the efficacy in HCC cell lines. To accelerate their future clinical use, possibly through drug-repurposing, we selected two old drugs to test in mice. Both drugs inhibited orthotopic liver tumor growth, suggesting that this bioinformatics approach could be a shortcut for developing anti-tumor strategies.
近年來以中草藥作為疾病(例如慢性肝臟疾病)替代療法的例子大幅增加,在先前的研究中,我們已經找到一種可治療肝硬化及肝癌的中草藥。為了將這些中草藥發展為治療用藥以及在實際的臨床上應用,我們做了更一進步的研究,但在這過程中,遇到了幾個瓶頸及挑戰,首先,中草藥的相關研究缺少系統化的科學驗證,而其來源品質及成分組成比例也不一致,除此之外,其中的作用機轉目前也了解的不夠透徹,因此,針對這些問題,我們利用connectivity map提供了一個可解決的方法。Connectivity map是一個儲存了大量基因表現圖譜的基因微陣列資料庫,可將疾病、基因表現及藥物三者做一個連結。先前我們已經利用connectivity map建立了一個利用肝癌病人基因表現圖譜來比對藥物的平台,我們的假設為藥物誘導的基因表現圖譜是可部分逆轉腫瘤不正常的基因表現圖譜,以這樣的理論為基礎,我們可以找到適合的藥物來進行治療肝癌,而參與在其中的分子,便可作為將來治療肝癌的標的。透過實驗,我們已經在許多肝癌細胞株中驗證了這樣的理論。為了加速藥物進入臨床的研究,我們選擇了其中兩種有效的老藥來進行動物實驗,實驗結果顯示這兩種藥物在動物實驗中都被發現可以抑制腫瘤的生長,由此可知,我們所設計的以基因表現圖譜為基礎的比對平台及藥物篩選實驗設計,確實可以發展為一種對抗癌症的治療方式。
